چکیده

Chromatin regulators (CRs) are essential upstream regulatory factors of epigenetic modification. The role CRs in the pathogenesis renal ischemia-reperfusion injury (IRI) remains unclear. We analyzed a bioinformatic analysis on differentially expressed chromatin regulator genes IRI patients using data from public domains. hub identified were used to develop risk prediction model for IRI, and their expressions also validated Western blot, qRT-PCR, immunohistochemistry murine model. examined relationships between infiltrating immune cells network explore drugs that target relevant downstream microRNAs. results machine learning methods showed five (DUSP1, GADD45A, GADD45B, GADD45G, HSPA1A) upregulated with key roles cell cycle, p38 MAPK signaling pathway, p53 FoxO NF-κB pathway. Two network, GADD45A GADD45B (growth arrest DNA damage-inducible protein 45 alpha beta), chosen They all good performance testing validation cohorts. Mice significantly expression within kidneys compared sham-operated mice. correlations plasmacytoid dendritic (pDCs) enrichment pathway based weighted gene co-expression (WGCNA) method. Candidate include beta-escin, sertraline, primaquine, pimozide, azacyclonol. dysregulation is related infiltration pDCs, may have therapeutic potential IRI.

Berberine Ameliorates Renal Functional Disorders and Pulmonary Tissue Injury Following Renal Ischemia/Reperfusion in Rats

Introduction: This study investigated the effect of berberine on renal dysfunction and histological damages of the lung induced by renal ischemia/ reperfusion at an early stage. Methods: There were four experimental groups of adult male rats (n=7). Seven days before induction of ischemia, the Ber+I/R group received oral (by gavage) berberine (15 mg/kg/day) while the I/R group received distil...

Amelioration of rat renal ischemia/reperfusion injury by L-Nil

Introduction: Ischemia/reperfusion (IR) injury involves a complex interrelated sequence of events. High levels of nitric oxide (NO) are generated with inducible form of nitric oxide synthase (iNOS) leading to the renal IR injury and glutathione (GSH) depletion. The present study was designed to investigate the effect of L-Nil (N6- (1-Iminoethyl)-L- lysine.hydrochloride), a selective inhibito...

Screening plasma miRNAs as biomarkers for renal ischemia-reperfusion injury in rats

BACKGROUND Acute kidney injury is a common clinical comorbidity and early diagnosis is crucial for improving prognosis, but there is still no ideal biomarker for early diagnosis. MATERIAL AND METHODS miRNA microarray was used for detecting miRNA in kidney subjected to renal ischemia-reperfusion injury 12 h after reperfusion. Real-time PCR was performed to validate the results of microarray. m...

Renal-associated TLR2 mediates ischemia/reperfusion injury in the kidney.

TLRs are conserved pattern recognition receptors that detect motifs of pathogens and host material released during injury. For unknown reasons, renal TLR2 mRNA is mainly expressed by tubular cells and is enhanced upon renal ischemia/reperfusion (I/R) injury. We evaluated the role of TLR2 in I/R injury using TLR2-/- and TLR2+/+ mice, TLR2 antisense oligonucleotides, and chimeric mice deficient i...

Combination Antioxidant Effect of Erythropoietin and Melatonin on Renal Ischemia-Reperfusion Injury in Rats

Objective(s): Renal ischemia reperfusion (IR) contributes to the development of acute renal failure (ARF). Oxygen free radicals are considered to be principal components involved in the pathophysiological tissue alterations observed during renal IR. The purpose of this study was to investigate the effect of co-administration of melatonin (MEL) and erythropoietin (EPO), potent antioxidant and ...
